Clonal Study of Murine Macrophages


Abstract
The murine homologous macrophage hybridoma cell lines were obtained by somatic cell fusion between Picibanil-elicited peritoneal macrophages and HAT-sensitive marine myelomonocytic cell lice, P388D1-S. The hybridoma cell lines that were grown in vitro for more than a year were confirmed by the expression of phagocytosis, adherence, monocyty specific nonspecific esterases, H-2 antigen, karyotypes, and other cell surface markers. In addition to the characterization of hybridoma cells. I investigated their ability to produce cytokines. After stimulation with LPS, supernatants of hybridoma clones showed IL-I and IL-i inhibitor activities. This study shows that homologous somatic cell fusion with marine macrophages can be used to define heterogeneous macrophage functions and gene expressions at the clonal level.
